| The Acclaim® 300 columns are a wide-pore line of C18 reversed-phase columns manufactured by for the high-resolution separation of proteins, peptides, and other biological macromolecules. The unique bonding chemistry of Dionex Acclaim columns show you what lies between the peaks, column after column. The Acclaim 300 was specifically designed for analysis of proteins and peptides, and features 3-µm silica particles for rapid analysis of complex protein digests. Because variations in both the raw silica and the ligand bonding chemistry and density significantly impact performance, each step of the process uses established procedures to maintain tight control. We start with ultrapure silica that contains minimal concentrations of metal contaminants, minimizing the tailing effects of residual, exposed silanol groups. Next, taking advantage of our unique bonding chemistries, we apply exhaustive bonding and end-capping techniques. These techniques result in stable bonding and columns that exhibit predictable reversed-phase separations with minimal secondary interaction mechanisms. Because of the stable bonding, only insignificant bleeding occurs, making these columns compatible with LC/MS applications. The packing procedures for the Acclaim 300 ensure maximum efficiencies. The Acclaim 300 can readily resolve proteins that differ by only a few amino acid residues. The Acclaim 300 phases have very high surface coverage, resulting in high-capacity columns that are stable between pH 2 and 8. The use of a 3-µm silica particle accelerates the diffusion of the mobile phase into the stationary phase, resulting in fast, high-resolution separations. Compared to 5-µm column packings, it is possible to increase the flow rate of the mobile phase and run shallower gradients on shorter columns to achieve the desired separation in a shorter time period. To meet the exacting needs of our customers, each Acclaim column is manufactured to stringent specifications to ensure the column-to-column reproducibility. Each column is shipped with a lot validation sheet showing the test results and specifications for the lot of bonded silica packed into the column and an individual column test chromatogram validating performance. The lot validation includes a chromatography-based metals test and tests for polar selectivity, hydrophobic steric selectivity, and base asymmetry. Production columns are individually tested for capacity and efficiency, and closely monitored for metals contamination. Reproducible column-to-column manufacturing, the use of ultrapure silica, and performance validation of every column prior to shipping, all ensure that the quality of your separations will be maintained month to month and year to year.
