CarboPac PA100 Column for Oligosaccharide Analysis

Characterization of oligosaccharides released from therapeutic glycoproteins is an important task for the biotechnology and pharmaceutical industries. Differences in glycoconjugate-derived oligosaccharides can be as subtle as differences in branching, linkage isomerism, and anomericity. These oligosaccharides can be challenging to separate by other methods, but are easily resolved by the CarboPac PA100. The CarboPac PA100 provides the flexibility to separate oligosaccharides into broad classes depending upon the degree of sialylation, while maintaining the resolution needed for routine analysis of these oligosaccharides based on linkage isomerism. Detection of these compounds at low-picomole levels has been optimized using pulsed amperometric detection (PAD). No sample derivatization is needed for detection. For researchers wishing to collect and further analyze the carbohydrate samples, the CMD may be used to desalt and reduce the pH of the samples following HPAE-PAD.

Oligosaccharides are also routinely determined in food and beverage products for a variety of purposes, such as quality control, verifying food labeling claims, establishing product authenticity, and monitoring fermentation processes. The CarboPac PA100 is an ideal column for detecting adulteration in natural fruit juices, or simply monitoring the levels of fermentable and nonfermentable sugars in beer production based on oligosaccharide profiling.

The CarboPac PA100 column is a high-resolution, strong, anion-exchange column developed for enhanced chromatography of oligosaccharides. The entire manufacturing process (including resin synthesis, amination, and packing of the columns) is carefully controlled to ensure that every CarboPac PA100 delivers reproducible performance.  CarboPac PA100 columns are tested with two isomers of N-acetylneuraminosyl-D-lactose to ensure lot-to-lot consistency.

Resin composition

8.5-µm-diameter ethylvinylbenzene/divinylbenzene substrate (55% cross-linking) agglomerated with 275-nm MicroBeadTM quaternary amine functionalized latex (6% cross-linked).
